Katarakt ist eines der häufigsten erblichen Probleme bei Australian Shepherds und sicherlich die häufigste vererbbare Augenerkrankung dieser Rasse. Typisches klinisches Zeichen ist die Trübung der Augenlinse, die zur Erblindung führt. Die primäre/vererbte Form der Krankheit tritt bei etwa 100 Hunderassen auf, aber trotz ihrer Häufigkeit ist die genetische Grundlage schlecht verstanden. Bis heute wurde nur das HSF4-Gen mit erblichen Katarakten bei Hunden in Verbindung gebracht und ist auch verantwortlich für die Entwicklung der Krankheit bei Australian Shepherds, Boston Terriern, Staffordshire Bull Terriern und Französischen Bulldoggen.

Die Vererbung der Krankheit ist relativ komplex aufgrund ihres autosomal dominanten Modus und der unvollständigen Penetranz, was bedeutet, dass nicht alle heterozygoten Tiere die Krankheit entwickeln. Die klinische Manifestation variiert erheblich. Pathologische Veränderungen entwickeln sich an verschiedenen Teilen der Linse, der Beginn der Krankheit variiert von der frühen Erwachsenenzeit bis zum Rand des Alters, und der Verlauf der Krankheit ist ebenfalls sehr variabel. Diese Fakten deuten darauf hin, dass andere noch unbekannte genetische und Umweltfaktoren zur Krankheitsentwicklung beitragen. Laut der wissenschaftlichen Literatur ist die Wahrscheinlichkeit, die Krankheit zu entwickeln, bei HSF4-heterozygoten Tieren 17-mal höher im Vergleich zu HSF4-clear Tieren. Der Test kann andere genetische Defekte nicht ausschließen, die möglicherweise an der Entwicklung der erblichen Katarakt bei Australian Shepherds beteiligt sind.

Bei Boston Terriern, Staffordshire Bull Terriern und Französischen Bulldoggen finden wir zwei Arten von erblichen Katarakten. Die früh einsetzende erbliche Katarakt (EHC) betrifft Hunde im Alter von wenigen Wochen, ist immer progressiv und bilateral und führt zu vollständiger Erblindung. Die spät einsetzende erbliche Katarakt (LHC) betrifft normalerweise Hunde über 3 Jahre und ist in ihrem klinischen Phänotyp, dem Alter des Auftretens, dem Verlauf und dem Grad der Sehbeeinträchtigung variabler. Die Mutation im HSF4-Gen wurde nur bei Hunden berichtet, die von EHC betroffen sind.

Vererbung: autosomal dominant (unvollständige Penetranz) - Australian Shepherd -;

autosomal rezessiv - Boston Terrier, Staffordshire Bull Terrier und Französische Bulldogge -.

Mutation: HSF4-Gen

Gentest: Die verwendete Methode zum Testen ist äußerst genau und ermöglicht eine vollständige Differenzierung zwischen betroffenen Tieren, Trägern und gesunden Hunden. Der Test kann in jedem Alter durchgeführt werden.

Australian Shepherd: Da die Krankheit autosomal dominant mit unvollständiger Penetranz ist, wird sie sich bei den meisten Tieren manifestieren, die mindestens ein mutiertes Gen tragen. Solche heterozygoten Tiere werden klinische Anzeichen zeigen, die ihre Lebensqualität beeinträchtigen. Mit der Absicht, die Krankheit auszurotten, ist es nicht ratsam, betroffene und Träger-Tiere zu züchten.
