Background
B7-H6, a glycosylated member of the B7 family of immune co-stimulatory proteins, is structured with a 238-amino acid extracellular domain containing one Ig-like V domain and one Ig-like C1 domain, a 21-amino acid transmembrane segment, and a 171-amino acid cytoplasmic domain featuring one ITIM, one SH2, and one SH3 motif. Both Ig-like domains undergo N-linked glycosylation. Notably, B7-H6 is expressed in various primary human tumors, such as leukemia, lymphoma, and gastrointestinal stromal tumors, but it is not constitutively expressed in normal tissues.
