Background
Phospho-α-Synuclein (Ser129) is the pathognomonic form of α-synuclein in which the serine at position 129 is stoichiometrically phosphorylated, a post-translational switch that converts the natively unfolded presynaptic protein into a sticky, aggregation-prone seed capable of nucleating Lewy bodies and Lewy neurites, propagating trans-synaptically between neurons, activating microglia and complement, and serving as the most specific biochemical correlate of dopaminergic neurodegeneration in Parkinson’s disease and dementia with Lewy bodies.
