De game heeft namelijk een nieuwe verhaallijn, nieuwe wapens, nieuwe monsters en uiteraard nieuwe omgevingen. Bovendien heb je het originele spel niet nodig om Ambition of Illuminus te spelen.

Het verhaal van de singleplayermodus gaat verder waar PSU ophield. Een nieuwe Human Fundamentalist Group, de Illuminus, verstoort het vredige Gurhal System en gooit de hele Gurhal Government overhoop. Met hun zelfgemaakte Guardian moeten spelers verschillende missies uitvoeren waarin ze bekende personen uit het eerste deel tegenkomen, zoals Ethan Waber.

Naast een singleplayermodus heeft PSU een uitgebreide MMO-multiplayermodus en die wordt met Ambition of Illuminus verder uitgebreid. Zo kunnen spelers elkaar nu ontmoeten in grote lobby's met kleurrijke decoraties en muziek. Ook kun je in lobby's naar een casino, waar je met roulette en speelkasten speciale items kunt winnen. Daarnaast kun je personages en wapens nu uitgebreider aanpassen dankzij de toevoeging van nieuwe wapens, uitrustingen, Photon Arts en kleding.
